I am wanting to set up my own online store, selling my tshirts. It would need to have a drop down menu, so that the customer can select what size they would like to purchase. I was wondering what you guys think is the best online store platform.

Most good ecommerce programs can accomodate what your need (different sizes, colors, etc).
Some ecommerce programs are free, some cost some $$.
Some are all-in-one (hosting, template, cc processing) - but those usually have design limitations and in the long run are more costly. Also be wary of proprietary ecommerce solutions (yahoo stores and others), you can not transfer your store to another web host if you wanted.

Personally I like the flexibility of http://www.magentocommerce.com/, however it can be a bit daunting at first, and professional help is usually needed to tweak things to your liking.

A simpler solution could be http://lemonstandapp.com/.

If you only plan to sell T-Shirts, I would take nhgnikole's suggestion for CafePress.

There are couple of eCommerce shopping sites available which would provide the basic functionality of having commerce website with basic payment gateway integration and listing your products online. Magento, OsCommerce and Drupal are popular ecomerce CMS. Along with selecting CMS you first need to register domain and buy hosting space.
