If you know anybody from the company your applying to, it would help if you ask them how they conduct interviews. At least you'll have an idea on what to expect.

Of course that is not always the case - it actually happened to me on my last job interview, but that was the first and only time so far. I would say, above all, understand what the job really requires and think long enough about it, why is it that you want it. If you are applying just because you want "a job" (not that there´s something wrong with it, everybody needs one, but...), it will eventually manifest itself through your words, you won´t sound convinced and unless they desperately need someone or your references are impeccable (but that should not be the case if you are just trying to land a job...), then they will not hire you. Settle for the position best suited for you and practice a little at home the usual questions. If you think you can make it, you probably will. You will come through as the right person for the job. Of course that does not guarantee 100% success, but what does in the end? There is no fail-safe approach to this matter.
